Almotriptan
Almotriptan is a selective 5-HT1B/1D receptor agonist used for the acute treatment of migraine attacks with or without aura in adults. It is a synthetic molecule that specifically binds to 5-HT1B and 5-HT1D receptors in the cerebral and meningeal blood vessels, activating them to reduce the pain and other symptoms of a migraine.
The mechanism of action involves the constriction of the dilated cerebral blood vessels, which helps in reducing the inflammation and nerve activation that cause migraine pain. It also inhibits the release of certain neurotransmitters, such as substance P and calcitonin gene-related peptide, that contribute to the pain and inflammation of migraines.
Almotriptan is available as tablets in dosages of 6.25 mg and 12.5 mg. The recommended starting dose is 6.25 mg, and it can be repeated once after 2 hours if the migraine returns. The maximum daily dose is 25 mg within a 24-hour period.
Almotriptan is not intended for the preventative treatment of migraines or for the management of chronic daily headaches. It should only be used when a clear diagnosis of migraine has been established.
