Sofosbuvir

Sofosbuvir is an oral antiviral medication used for the treatment of chronic hepatitis C virus (HCV) infection. It is a nucleotide analogue that inhibits the HCV NS5B RNA-dependent RNA polymerase, thus blocking the viral replication process.
Sofosbuvir is commonly used in combination with other antiviral drugs, such as ledipasvir, daclatasvir, or ribavirin, to increase its effectiveness and reduce the risk of treatment resistance. The specific combination and duration of therapy depend on the genotype of the HCV strain, the presence of liver fibrosis, and the patient’s overall health condition.
The primary indication for sofosbuvir-based regimens is the treatment of chronic HCV infection in adults with compensated liver disease, including those with cirrhosis. Sofosbuvir has demonstrated high cure rates across various HCV genotypes and patient populations, including those with HIV coinfection, advanced liver disease, or prior treatment failure.
Sofosbuvir is generally well-tolerated, with a favorable safety profile. The most common side effects reported in clinical trials include fatigue, headache, and nausea. Sofosbuvir is contraindicated in patients with severe renal impairment or decompensated liver disease.
